AB OBJECTIVE: The aim of the study was to look at the concentration of
Natural Killer (NK) cells in two risk groups infected with HIV 50
intra-venous drug users were analysed and 50 homo/bisexual men were the
second cohort. METHOD: Blood was incubated with dual labelled monoclonal
antibodies as follows- 1. lymphocyte gating MoAb CD45/14 2. isotype
control and 3. T-cell and NK marker CD3/16 + 56 (Becton Dickinson) The
red blood cells were lysed and the remaining cells analysed by flow
cytometery--FACScan (Becton Dickinson). RESULTS: The average of the 50
IVDU (38 male:12 female) was 6.8% positive staining for CD16 + 45 in the
lymphocyte 'gate'. In the sexually aquired cohort the mean was 15.7% of
similar cells. DISCUSSION: NK cells are reported to exhibit minimal
specificity and have no memory. Their cell lineage is not known. Also
they are non-adherent and non-phogocytic. The above results show a
marked difference in the two cohorts. Differing viral strains may be the
agent?
